  • Solid podcast

The guy needs to stop saying "um" and "uh" so much between his thoughts.

  • Good information, bad delivery

Often times a podcast with very good information, but quite poorly delivered. As someone who regularly conducts market research interviews via telephone and web, I’m very familiar with reading through far too many colloquialisms, filler words, and trite expressions. Words and phrases such as “right “and you know” “um” “and” “uh” make it painful to listen to in any regular basis.

Talking Points

Recent interactions between the hosts and their guests.

Inside Step Pharma’s Pan‑Cancer Ambition
Q: How do you envision expanding CTPS1 inhibitor use across cancer types?
We start with lymphomas to establish efficacy, then move to solid tumors by selecting patients whose tumors have CTPS2 loss, creating a biomarker-driven pan-cancer approach. In parallel, we are exploring a two-product strategy with distinct dosing regimens for solid tumors versus essential thrombocythemia to maximize patient benefit and market potential.
Inside Step Pharma’s Pan‑Cancer Ambition
Q: What has been the biggest clinical challenge, and how has the team addressed it?
The major challenge has been identifying and managing the unintended platelet suppression. Through extensive PK-PD modeling and trial iterations, we found that a one-week-on/one-week-off dosing schedule mitigates this platelet effect while maintaining anti-tumor activity, supported by cumulative clinical data showing no recurrent adverse events.
Inside Step Pharma’s Pan‑Cancer Ambition
Q: Could you explain the core mechanism Step Pharma is leveraging with CTPS1 and why it is cancer-selective?
CTPS1 inhibition depletes CTP, blocking DNA and RNA synthesis. Normal cells rely on CTPS2 when CTPS1 is blocked, while many cancer cells, due to their rapid proliferation and specific dependencies, become vulnerable to CTPS1 inhibition. In lymphocytes, CTPS1 loss severely impairs proliferation, but other tissues remain largely unaffected, enabling targeted anti-cancer activity with a favorable safety profile.
No Matter What: How Israel Is Positioning Itself as a Global Pharma Innovation Hub
Q: What are the three structural gaps in Israel's ecosystem, and how does AION Labs plan to address them?
The three gaps are experienced leadership, early-stage capital, and translational capabilities. AION Labs plans to address them by leveraging pharma partners for mentoring and validation, providing initial funding and access to venture networks, and building translational platforms to bridge academia and industry.
Why Local Matters: A Leadership Perspective on Community-Based Trials
Q: What needs to happen for community-based access to become a true industry standard?
The industry needs to take a broader view of trial design and invest in community engagement over a long period to normalize community-based clinical research.
