  • Great content, can’t understand

    Great selection of papers

The lady with the accent is very difficult to understand. She needs to speak slower, at times she speeds up and I cannot follow what she is saying. I am speaking for a lot of people probably too

Please slow it down and also try to speak about limitations in the studies more frequently

  • Love love love this podcast.

The content is solid and I garner new insight from every episode. Even though I focus mostly on immunooncology, I appreciate the wide diversity of the topics covered as they expand my thinking about cancer into directions I might not have gone otherwise. I look forward to long road trips catching up on old episodes.

Talking Points

Recent interactions between the hosts and their guests.

Ep. 131: “Lymphocyte Development” Featuring Dr. Alfred Singer
Q: Could you start with a little overview about your early research, and how our understanding of the role of the thymus in T-cell development evolved from the 70s or 80s to today?
Dr. Singer describes how thymus-derived progenitors become T cells through signaling events, the emergence of double-positive thymocytes, and the selection processes that determine CD4 versus CD8 fates, emphasizing that signaling duration and co-receptor interactions shape fate decisions.
Ep. 129: Live from IMMUNOLOGY2026TM: Emerging Immunotherapies for Solid Tumors
Q: How is the field approaching safety and toxicity management in engineered cell therapies?
Approaches include implementing safety switches, regulating cytokine production (e.g., limiting IL-12 activity), developing targeted delivery to tumor sites, and using in vivo manufacturing methods that allow more controlled, transient expression while monitoring for genotoxicity and off-target effects.
Ep. 129: Live from IMMUNOLOGY2026TM: Emerging Immunotherapies for Solid Tumors
Q: What are the main obstacles to achieving durable responses in solid tumors with CAR T cells?
Barriers include the physical stromal barrier that hinders infiltration, tumor antigen heterogeneity leading to relapse, and the risk of on-target/off-tumor toxicity; strategies include multi-antigen targeting, local delivery, and combining CAR T with agents that remodel the microenvironment.
Ep. 126: “Functional Oncogenomics” Featuring Dr. Daniel Peeper
Q: What are the main pathways that influence tumor sensitivity to immunotherapy according to the guest?
Main pathways include the antigen presentation machinery, interferon gamma signaling (with a complex impact), TNF signaling, and autophagy, with TNF and interferon gamma showing the strongest and most reproducible effects across in vitro and in vivo settings.
Ep. 126: “Functional Oncogenomics” Featuring Dr. Daniel Peeper
Q: What makes functional genetics different from traditional genomic screens?
Functional genetic screens test causality by knocking out (or activating) genes across the genome and observing direct effects on cellular phenotypes, such as tumor cell sensitivity to T-cell killing, which helps identify targets with real mechanistic roles rather than mere correlations.
