  • Oncology Brothers: Setting a New Standard in Medical Education

The Oncology Brothers have become a go-to in my week. They take the constant firehose of oncology data—new FDA approvals, shifting algorithms, and “practice-changing” abstracts—and turn it into something practical I can actually use in clinic the next day. The conversations feel like sitting down with smart colleagues who care about evidence, clarity, and real-world decision making, not just reciting trial slides. It’s exactly the kind of thoughtful, high-yield education our field needs more of.... more

Thank you so much for putting this together! Love the updates of ongoing research! I am the primary caretaker for my 79 year old dad who was diagnosed 6/24 with stage IV CRC MSS KRAS G13D TMB-H, so love your GI updates from ASCO ‘25 and subcutaneous nivo, which we are considering. Hope you do more like bot/bal and neodadjuvant single cycle pembro. Thanks for taking the time to do this!!

Talking Points

Recent interactions between the hosts and their guests.

RASolute 302 Pancreatic Cancer Study with Daraxonrasib at ASCO 2026 – Dr. Brian Wolpin
Q: What were the key side effects, and how should clinicians manage them in practice?
The most common toxicities were rash, stomatitis, diarrhea, and nausea. Management includes prophylactic dermatology measures, antibiotics like doxycycline, topical steroids, sun protection, antiemetics, and dose modifications when needed.
RASolute 302 Pancreatic Cancer Study with Daraxonrasib at ASCO 2026 – Dr. Brian Wolpin
Q: What makes Daraxonrasib distinct from prior RAS inhibitors in terms of mechanism?
Daraxonrasib is a RAS-ON inhibitor and a multi-selective agent that binds both mutant alleles like G12D/V/R and wild-type RAS, acting through a molecular glue mechanism with cyclophilin A to block signaling.
RASolute 302 Pancreatic Cancer Study with Daraxonrasib at ASCO 2026 – Dr. Brian Wolpin
Q: Could you explain who was eligible for the trial and how broad the enrollment was with respect to RAS status?
The trial enrolled metastatic pancreatic cancer patients who had one prior chemotherapy; RAS status documentation was required but not mandatory for inclusion, making the population broadly representative and including patients with and without a documented RAS mutation.
Challenging Cases in Relapsed/Refractory Acute Myeloid Leukemia (AML): Dr. Joshua Zeidner
Q: What makes Revumenib a preferred second-line option after relapse post-transplant in KMT2A rearranged AML?
Revumenib demonstrates higher response rates (around 60%+ overall) and meaningful remissions, making it a strong bridging therapy to longer-term strategies like transplant, with tolerability that supports outpatient or less intensive management.
Managing Toxicities of BRAF-MEK Inhibitors in Non-Small Cell Lung Cancer – Dr. Melissa Johnson
Q: What are the two main BRAF-MEK inhibitor combinations discussed, and how do their efficacy data compare?
The two combinations are Dabrafenib plus Trametinib, with earlier data showing meaningful responses and longer OS in melanoma and NSCLC, and Encorafenib plus Binimetinib, which recent data demonstrate longer progression-free survival and favorable overall survival in NSCLC; the choice depends on tolerability and patient-specific considerations.
