Fungal cultures are time consuming but highly beneficial when samples are viable and collected properly; outsourcing to trained TVMDL staff can relieve practitioners of the logistical burden and improve diagnostic accuracy for their patients.

Q: So why would we want to do a culture over, say, PCR testing?
Culture provides information on spore viability and serves as a reliable follow-up method to monitor treatment progress, even though PCR offers faster initial results. Cultures can be held for several weeks to confirm viability and guide decisions about when an animal is truly clear of infection.

Q: How should veterinarians interpret the results once they get them?
A positive AGID or CF result indicates exposure to the fungus but does not confirm active disease; rising antibody titers over time can suggest active infection, and results must be interpreted alongside clinical signs and imaging findings to determine the likelihood of active disease or relapse.

Q: What specimens are required for all those types of tests?
For all the tests discussed, collect one to two milliliters of serum from a clotted blood sample in a non-additive tube like a plain red-top or a serum separator tube, then carefully transfer clear serum to a new tube to minimize hemolysis, especially in hot Texas weather.

Q: Kalyn, walk us through what the diagnostic tests we have available at TVMDL are?
TVMDL offers several tests including AGID for detecting antibodies to Coccidioides immitis, CF to measure antibody levels and gauge severity, and multiple fungal panels that combine these AGID tests with additional pathogens to broaden surveillance. The panels include combinations with Aspergillus and Cryptococcus species to provide a broader serologic profile, with CF used to monitor antibody titers over time for disease activity.
