When listening to a podcast for the first time, I start at the beginning. They do often get better over time as the podcaster gains experience. This podcaster gets positive points for clearly stating that she isn’t an expert, that she is learning along with the audience. I definitely allow a lot of slack for pronunciation, scientific terms are often difficult to pronounce or have >1 accepted pronunciation.

But, while listening to episode #2, I turned it off after the 5th time the podcaster said ... more

  • Frustrating

    This podcasts interviews

lots of great experts with a deep knowledge but always frustrates me because the technical stuff is never really addressed in any depth. With some genetic knowledge I repeatedly find it a missed opportunity, as the questions I want to ask are omitted. If it has a huge lay audience that’s fine, but not for me.

Talking Points

Recent interactions between the hosts and their guests.

#390 Prince, Mayte Garcia, and Their Son Amiir’s Pfeiffer Syndrome Type 2 Story
Q: How did the decision to forego prenatal testing shape the pregnancy and Amir's early days?
The host discusses that the decision was deeply personal, balancing faith and uncertainty, and allowed the parents to focus more on Amir's immediate life with love and time rather than medical terminology.
#389 From Natural History to Gene Therapy: The Future of Danon Disease Research
Q: What are the key eligibility criteria for the interventional Lamplight trial?
Eligibility focuses on males aged eight years or older with a confirmed pathogenic or likely pathogenic LAMP2 mutation, left ventricular hypertrophy with preserved ejection fraction, and New York Heart Association class 2 or 3, excluding those with other organ transplants or significant cardiovascular support needs.
#389 From Natural History to Gene Therapy: The Future of Danon Disease Research
Q: Can you explain why gene therapy is a potential treatment modality for Danon disease, especially with the associated cardiomyopathy?
Gene therapy aims to restore the function of the damaged LAMP2B gene by delivering a normal copy to cardiac myocytes, with the goal of producing the missing protein and slowing or reversing cardiac deterioration, thereby addressing the root cause of the disease.
#386 Achondroplasia Beyond Height: Managing Lifelong Medical Needs
Q: How has the treatment landscape changed in the last 5–10 years?
There is now more data and treatment options being discussed earlier with families, but careful, shared decision-making and accurate information are essential to avoid misinformation.
#385 Inside ACMG 2026: How AI and New Tools Enhance Genome Sequencing and Equity
Q: What evidence exists that AI can handle complex scenarios like dual diagnoses or variants of uncertain significance?
Studies show AI can identify both known disease-genic variants and variants of uncertain significance, sometimes flagging dual diagnoses and helping prioritize which variants to pursue for reporting, thereby expanding diagnostic possibilities beyond simple single-diagnosis cases.
