We started from the idea that signaling pathways underlying endothelial response to flow may be repurposed in disease; FOXO1 emerged because it integrates metabolic cues with inflammatory and growth-regulatory programs, and our data showed its activity changes with disturbed flow in endothelial cells, suggesting it could couple hemodynamics with transcriptional control relevant to atherosclerosis.
