Q: How did you handle the dose levels and what did you observe about plasma levels and responder status?
Higher plasma concentrations were associated with responders, and preselection based on metabolism helped manage adverse events. Despite this, adverse events did not differ significantly between randomized groups, supporting genotype-guided dosing as a potentially beneficial strategy.

Q: Why did you go for Nortriptyline? Why not amitriptyline or another neuromodulator?
Nortriptyline was chosen because it has fewer anticholinergic adverse events than amitriptyline, and its simpler metabolism (single enzymatic step) facilitates a more precise, personalized dosing approach through preselection based on genotype.
